ABSTRACT

OBJECTIVE@#To investigate the effect of sputum ubiquitin ligase (Cbl-b) gene known-down on the cytotoxicity of H9 T lymphocytes against human laryngeal squamous cancer Hep-2 cells and explore the underlying mechanism.@*METHODS@#CD4 T lymphocytes isolated from 12 patients with laryngeal squamous carcinoma and 12 healthy individuals were examined for Cbl-b mRNA expressions using RT-PCR. H9 T lymphocytes cultured in 96-well plates were transfected with Cbl-b siRNA via liposomes followed by treatment with an anti-IL-2 monoclonal antibody, with H9 T lymphocytes transfected with a scrambled sequence as the negative control. The expressions of Cbl-b mRNA and protein in the cells were detected using real-time fluorescent quantitative PCR and Western blotting, respectively. The killing effect of the treated T lymphocytes against Hep-2 cells was assessed using the cell counting kit (CCK-8). The positive expression rates of CD69 and CD25 on the surface of H9 T lymphocytes were determined using flow cytometry, and the levels of interleukin-2 (IL-2) and interferon-gamma (INF-γ) in the culture supernatants of H9 T lymphocytes were detected with ELISA.@*RESULTS@#The CD4 T lymphocytes from patients with laryngeal squamous carcinoma showed significantly increased Cbl-b mRNA level compared with those from healthy individuals ( < 0.05). Transfection of H9 T lymphocytes with Cbl-b siRNA significantly reduced the expression levels of Cbl-b mRNA and protein ( < 0.05), which were not significantly affected by subsequent treatment of the cells with the anti-IL-2 antibody (>0.05). At different target-effector ratios, the Cbl-b siRNA-transfected cells showed significantly higher Hep-2 cell killing rates and higher positivity rates of CD69 and CD25 expressions than the blank and negative control cells and the cells with both Cbl-b siRNA transfection and anti-IL-2 treatment ( < 0.05). Cbl-b silencing in H9 T lymphocytes resulted in significantly increased levels of IL-2 and INF-γ in the supernatant as compared with those in the blank and negative control groups ( < 0.05).@*CONCLUSIONS@#Cbl-b gene silencing effectively enhances the killing effect of H9 T lymphocytes against Hep-2 cells probably as the result of enhanced IL-2 secretion and T lymphocyte activation.

ABSTRACT

Objective To analyze the immunization coverage of 23-valent pneumococcal polysac-charide vaccine(PPV23)in a large population in Guangzhou and to evaluate its safety by analyzing the ad-verse events following immunization(AEFI)reported to the passive surveillance system. Methods Immu-nization data of PPV23 in Guangzhou from 2010 to 2015 were collected from Information Management System of Biological Products and the Information System of Immune Programming of Guangzhou. AEFI reported to the AEFI Information System during 2010 to 2015 was collected for safety evaluation. The collected data were analyzed by using descriptive methodology. Results A total of 621 059 doses of PPV23 were pre-scribed in Guangzhou from 2010 to 2015. Most of the recipients were children younger than 10 years old,ac-counting for 79. 44% . Only 9. 38% of the subjects received PPV23 were older than 60 years. A total of 243 AEFI cases were reported at a rate of 39. 13 cases per 100 000 doses,among which 199 cases(32. 04 / 105 ) showed minor vaccine reactions,25 cases(4. 03 / 105 )occurred adverse events,16 cases(2. 58 / 105 )de-veloped coupled diseases and 3 cases(0. 48 / 105 )were classified as psychogenic reactions. No rare adverse reactions were observed. Conclusion The majority of people immunized with PPV23s in Guangzhou were children,while the immunization coverage among the elderly was relatively low. PPV23 was safe for vaccina-tion as the reported AEFI cases were similar to that of other vaccines.

ABSTRACT

Objective To explore the influence of porcine esophageal variceal pressure on complete ligation for polycyclic ligator in vitro.Methods The experimental porcine venous vessels were selected to make the models of different venous pressure,which were divided into 3 groups according to the preset pressure range,P1 group 25-30 cmH2O(1 cmH2O =0.098 KPa),P2 group 35-40 cmH2O,P3 group 45-50 cmH2O.Ligation effects of each group were analysed.Results There were 75 complete ligations (47.77%,75/157) in group P1,41 in group P2 (32.28%,41/127),and 29 in group P3 (23.58%,29/123).There were significant differences among the three groups (x2 =19.558 5,P =0.000 6).Conclusion The higher the variceal pressure is,the worse the effect of ligation is.Variceal pressure could predict the effect of endoscopic ligation,and is helpful for the choice of endoscopic treatment.

ABSTRACT

Objective To explore the influence of different pressures and diameters in vitro on complete ligation degree of esophageal varices based on the principle of LDRf typing.Methods According to the different preset venous pressure,the experimental porcine veins were used to establish vascular pressure models and divided into three groups (group A 25-30 cmH2O,group B 35-40 cmH2O,group C 45-50 cmH2 O).The porcine esophagus was selected to establish models of esophageal varices,and divided into three groups according to the diameter of esophageal varices (group D1 0.4-1.0 cm,group D2 1.0-1.5 cm,group D3 1.6-2.0 cm).Influence of pressure and diameter on complete ligation degrees was evaluated by univariate and multivariate analysis.Results Group A completed 18 ligations (56.25%,18/32),group B 12 ligations (37.50%,12/32),and group C 11 ligations (33.33%,11/33),which were not significantly different by pressure (x2 =3.6126,P =0.0573).Group D1 completed 35 ligations (94.59%,35/37),group D2 6 ligations(16.67%,6/36),and D3 completed no ligation (0%,0/24),which were significantly different by diameter (x2 =38.0014,P =0.0000),indicating that the diameter of the varices exerted greater influence on the ligation degree.The logistic regression analysis showed that variceal pressure and diameter were the independent factors for complete ligation degree (P =0.000).Conclusion Complete ligation is effective and safe when the variceal diameter is 0.4-1.0 cm,or variceal pressure is 25-30 cm H2O.Endoscopic treatment for varices with the guide of LDRf typing is scientific and feasible.

ABSTRACT

Objective To study the feasibility of LDRf classification for gastrointestinal tract ectopic varices (EcV) outside the esophagus.Methods Data of 914 patients with gastrointestinal EcV were classified by LDRf and analyzed for EcV location (L),vascular diameter (D),and risk factor (Rf).The etiology of the portal hypertension (PH) was determined,and the patients were treated and followed up.Results The EcVs were located in duodenum of 198 cases,in jejunum and ileum of 93,in bile duct of 105,in colon of 65,and in rectum of 453.Diameters of blood vessels of EcV varied from 0.3 to 3.5 cm.PH causes were cirrhosis with portal hypertention in 630 patients (68.9%),in which 3 were autoimmune cirrhosis and 3 were portal spongiform liver disease (0.6%).Combined esophageal and gastric varices were found in 252 cases (27.6 %),including 4 cases (0.5%) of splenectomy.Various treatments were applied in 315 patients,including endoscopic tissue adhesive injection in 43,endoscopic sclerotherapy in 76,endoscopic ligation in 74,interventional treatment in 52,and surgical laparotomy in 70.A total of 19 patients died of variceal bleeding.The patients were followed up for 13 to 36 months,no varices relapsed,and 1-year survival rate was 100%.Conclusion LDRf classification,simple,applicable,standardized,is suitable for the whole gastrointestinal varicose veins.

ABSTRACT

ObjectiveTo observe the change of cardiac function after acute myocardial infarction (AMI) in rabbit model,and to study MRI characteristics of left ventricular remodeling (LVR).Methods Forty-five japanese white rabbits underwent chest-opening coronary artery ligation surgery to obtain rabbit myocardial infarction model The animals were scanned on a 1.5 T MR scanner ( GE Healthcare,Chalfont St.Giles,UK) at six time-points as surgery,1,2,4,6,8 weeks after surgery. Cardiac function parameters were measured,including left ventricular end diastolic volume ( EDV ),stroke volume ( SV ),ejection fraction (EF),peak ejection rate ( PER ) and peak firing rate (PFR).At each time point,5 rabbits were randomly selected and performed re-thoracotomy to obtain heart specimen.Each specimen was examined by MRI with the diffusion tensor imaging ( DTI ).The value of ADC and fractional anisotropy (FA) were collected. Cardiac function data sets of different time points were analyzed using repeated measures data of ANOVA.The FA and ADC values of infarction myocardium set of different time points or different parts of the myocardium were analyzed using one-way ANOVA. The average was compared statistically between two adjacent groups using LSD-t test. Results Left ventricular EDV increased in progress with the time.It was increased from ( 2.21 ± 0.35 ) ml preoperatively to ( 3.15 ± 0.57 ) ml 8-week postoperatively.But the difference was not statistical significant ( F =1.384,P =0.220).EF was decreased from(57.31 ± 11.11 )% to( 34.71 ± 8.72 )%.It dropped significantly and the difference was statistically significant (F =27.134,P =0.001 ).EF showed a downward trend with the increase of EDV.By linear regression analysis,an equation was set up with y =- 5.58x + 57.7 ( F =8.855,P =0.005 ).On the other hand,PER showed a progressive decline from (27.31 ± 13.06) ml/s to ( 17.31 ± 6.41 ) ml/s ( F =2.105,P =0.037 ),and PFR decreased from ( 23.79 ± 14.15 ) ml/s to ( 12.07 ± 6.91 ) ml/s ( F =2.366,P =0.011 ).FA value decreased from 0.442 ±0.093 to 0.107 ±0.037( F =12.61,P =0.001 ),and ADC valueshowed upward trend from (5.07 ± 1.15) ×10-4 mm2/s to(6.39 ±0.78) ×10-4 mm2/s (F=4.166,P =0.022 ).FA values of infarct,adjacent and remote region were 0.201 ± 0.049,0.316 ± 0.127 and 0.323 ± 0.117 respectively( F =3.896,P =0.004 ),and the ADC values in these regions were (6.19 ± 1.78 ) ×10 -4,(5.44 ± 2.63 ) × 1 0 -4,(5.29 ± 2.02 ) × 10 -4 mm2/s respectively ( F =3.248,P =0.018 ).FA and ADC values were significantly different between the infarct region and adjacent region ( t =7.327,P =0.001 ;t =3.292,P =0.005,respectively),but there was no significantly different between adjacent region and remote region ( t =1.024,P =0.129 ; t =1.467,P =0.164,respectively ).ConclusionsMRI measurement of parameters of cardiac function can be used to monitor the process of left ventricular function remodeling after AMI.The process of micro-structural remodeling of myocardium can be reflected by DTI.MRI provides a feasible imaging modality for LVR after AMI.

ABSTRACT

Objective To investigate the changes of carotid, lipidemia and fibrinogen in type 2 diabetes mellitus patients complicated with cerebral infarction (DCI). Methods 58 cases of DCI (DCI group) were enrolled in this study, and 36 cases of cerebral infarction without diabetes mellitus served as control group. The location, number and nature of carotid atherosclerotic plaque and inner diameter of carotid artery were assessed by Doppler ultrasonography in all the patients, as well as blood lipid, blood glucose and fibrinogen were evaluated.
